Angiogenesis is defined as the development of new blood vessels that form from pre-existing vasculature. Angiogenesis is a vital process that occurs during normal development, growth, and wound healing. However, it is also implicated in the growth of cancer, through the tumor’s ability to secrete chemical signals that stimulate angiogenesis. Angiogenesis is important in cancer. Tumors must have a viable blood supply to enable them to grow to a significant size as the tumor’s population of rapidly dividing cells require oxygen and important nutrients. An imbalance between cellular oxygen consumption and oxygen availability can stimulate hypoxic cancer cells (HCCs) to induce angiogenesis. The central role pro-angiogenic factors play in tumor-induced angiogenesis means that they are an attractive therapeutic target for fighting cancer. There are a number of ‘signals’ that lead to the release of pro-angiogenic factors, one of the strongest being hypoxia. The best characterized pro-angiogenic factor is vascular endothelial growth factor (VEGF), a homodimeric glycoprotein that binds two receptors found on the surface of vascular endothelial cells. VEGF can mediate angiogenesis in cancer through its up-regulation, via a variety of growth factors and by hypoxia. The cellular adaptive response to hypoxia is regulated by hypoxia-inducible factor (HIF), this transcription factor can induce the expression of VEGF along with many other pro-angiogenic factors.
